Indications

  • Treatment of skin and soft tissue infections
  • Treatment of respiratory tract infections
  • Treatment of osteomyelitis, enteritis, endocarditis, urinary tract infection, meningitis, septicaemia
  • Prophylactic agent for major surgical procedures

Pharmacology

  • Active against Gram-positive organisms
  • Little activity against Gram-negative bacilli
  • Inhibits the formation of bacteria cell wall
  • Resistance to hydrolysis by penicillinase
  • Bactericidal antibiotic
  • Kills bacterial cell wall by interfering with peptidoglycan synthesis

Dosage

  • Oral administration for adults: 250 mg four times daily
  • Dosage may be doubled in severe infections
  • Parenteral administration for adults: 250 mg-1 g four times daily
  • Oral doses should be administered 1 hour before meal

Interaction

  • Concurrent use may result in increased level of Halopen in blood for prolonged period

Contraindications

  • Contraindicated in penicillin hypersensitive patients

Side Effects

  • Common side effects of gastrointestinal tract such as nausea, vomiting, diarrhea, dyspepsia
  • Other side effects include rashes, urticaria, purpura, fever, interstitial nephritis, hepatitis, and cholestatic jaundice

  • US FDA Pregnancy Category B
  • Caution advised during pregnancy and lactation

Precautions & Warnings

  • Use with caution in patients with evidence of hepatic dysfunction
  • Use with caution in patients with an allergic diathesis
  • Reduced dose or extended dose interval in severe renal failure

  • Penicillinase-resistant penicillins

Reconstitution

  • Can be used in other routes in conjunction with systemic therapy
  • Administered by intraarticular and intrapleural injection
  • Administered by inhalation using a nebulizer
