Indications

Pharmacology
- Dihydropyridine calcium-channel blocker
- Vasodilation
- Reduced peripheral resistance
- Increased coronary blood flow
- Reflex increase in coronary heart rate
Dosage & Administration
- Usual dose: 5 mg once daily
- Maximum dose: 10 mg once daily
- Lower dose for elderly
- May be taken without regard to meals
Interaction
- Potentially hazardous interactions with p-blockers
- No interaction between Amlovas and Digoxin
- No interaction between Amlovas and Cimetidine
- No significant alteration of Warfarin effect
- Food does not alter absorption of Amlovas
Contraindications
- Hypersensitivity to dihydropyridine derivatives
- Pregnancy

Use in Special Populations
- Dosing recommendations for children with hypertension
- No known effect of amlodipine on blood pressure in patients under 6 years of age
- Normal dosage regimens recommended in the elderly
- Normal dosage recommended for renal impairment
- Caution in dose selection for hepatic impairment
Overdose Effects
- Excessive peripheral vasodilatation
- Reflex tachycardia
- Marked and prolonged systemic hypotension
- Active cardiovascular support required
- Elevation of extremities
- Attention to circulating fluid volume and urine output
- Benefit of intravenous calcium gluconate
- Possible use of gastric lavage
- Benefit of charcoal up to 2 hours after administration
- Dialysis unlikely to be beneficial
