Indications

Pharmacology
Amoxicillin is a broad spectrum penicillin effective against a wide range of bacteria. It inhibits cell wall biosynthesis, is stable in gastric acid, and rapidly absorbed after oral administration.
Dosage
- Adult:
- 250 mg three times daily
- Up to 500 mg three times daily for severe infections
- Children:
- 125 mg three times daily
- Up to 250 mg three times daily for severe infections
- Specialized Dosing:
- 3 gm every 12 hours for severe respiratory infection
- 1 g three times daily for otitis media in adults
- 40 mg/kg body weight daily in 3 divided doses for otitis media in children (max. 3 g daily)
- 500-1000 mg three times daily for pneumonia
- 3 gm for dental abscess, repeated after 10-12 hours
- 3 gm for urinary tract infections, repeated after 10-12 hours
- 2-3 gm single dose for gonorrhoea with Probenecid 1 gm
Administration
Reconstituted suspension can be administered by adding it to milk, fruit juice, or water and should be taken immediately.
Interaction
Concurrent use with Probenecid may increase and prolong blood levels. Can affect gut flora and lower estrogen reabsorption.
Contraindications
Contraindicated in penicillin hypersensitive patients.
Side Effects
- Mild and transient side effects
- May include diarrhea, indigestion, occasionally rash
- Rare reports of pseudo-membranous colitis
Pregnancy & Lactation
Amoxicillin is category B in pregnancy. Caution should be exercised during lactation as it is excreted in human milk.
Precautions & Warnings
Superinfections with mycotic or bacterial pathogens should be kept in mind during therapy. Appropriate therapy should be instituted if superinfections occur.
