Pharmacology
- Broad spectrum penicillin
- Effective against Gram-positive and Gram-negative bacteria
- Inhibition of cell wall biosynthesis
- Stable in gastric acid
- Rapid absorption after oral administration
- Widely distributed in body tissues
Dosage
- Adult dosage: 250mg three times daily
- Increasing dosage for severe infections
- Children dosage: 125mg three times daily
- Severe or recurrent purulent respiratory infection: 3g every 12 hours
- Otitis media: 1g three times daily for adults
- Otitis media: 40mg/kg body weight daily in 3 divided doses for children
- Pneumonia: 500-1000mg three times daily
- Dental abscess: 3g repeated after 10-12 hours
- Urinary tract infections: 3g repeated after 10-12 hours
- Gonorrhoea: Single dose of 2-3g with Probenecid 1g
- Renal impairment may require reduced dosage
Administration
Reconstituted suspension can be administered with milk, fruit juice, or water
Interaction
Concurrent use with Probenecid may increase and prolong blood levels
Contraindications
Contraindicated in penicillin hypersensitive patients

- Mild and transient side effects
- Diarrhoea
- Indigestion
- Rash (occasional)
- Pseudo-membranous colitis (rare)
Pregnancy & Lactation
- US FDA pregnancy category B
- Use during pregnancy only if clearly needed
- Excreted in human milk
- Caution advised during lactation
Precautions & Warnings
Superinfections with mycotic or bacterial pathogens should be considered

Reconstitution
- Intramuscular reconstitution: Add 2.5 ml water for injection to Amoxycillin 500 mg injection vial
- Intravenous reconstitution: Dissolve in 10 ml water for injection
