Indications
- Essential hypertension
- Angina pectoris
- Vasospastic angina
Pharmacology
- Calcium-channel blocker
- Vasodilation
- Reduced peripheral resistance
- Increased coronary blood flow
- Myocardial oxygen supply
Dosage & Administration
- Usual dose 5 mg once daily for hypertension
- Usual dose 5-10 mg for angina
- May be taken without regard to meals
Interaction
- Potentially hazardous interaction with p-blockers in patients with impaired left ventricular function
- No interaction with Digoxin
- No interaction with Cimetidine
- No significant alteration of Warfarin effect
- Food does not alter absorption
Contraindications
- Hypersensitivity to dihydropyridine derivatives
- Pregnancy

- Flushing
- Headache
- Hypotension
- Peripheral edema
- Gastrointestinal disturbances
- Increased micturition frequency
- Lethargy
- Eye pain
- Mental depression
- Ischaemic chest pain
- Excessive fall in blood pressure

- Abnormalities in liver function
Pregnancy & Lactation
- Pregnancy Category C
- Use only if potential benefit justifies potential risk to fetus
- Discontinue nursing while administered
Precautions & Warnings
- Take with caution in patients with hepatic impairment, pregnancy, and breastfeeding
Use in Special Populations
- Different doses for children with hypertension
- Normal dosage regimens recommended for elderly
- Normal dosage recommended for renal impairment
- Caution in dose selection for hepatic impairment
Overdose Effects
- Excessive peripheral vasodilatation
- Reflex tachycardia
- Marked systemic hypotension
- Management with cardiovascular support and close monitoring
- Use of vasoconstrictors and intravenous calcium gluconate
- Gastric lavage
- Consideration of charcoal if within 2 hours of administration
