Indications
- Essential hypertension
- Angina pectoris
- Vasospastic angina
Pharmacology
- Calcium-channel blocker
- Vasodilation
- Reduced peripheral resistance
- Increased coronary blood flow
- Myocardial oxygen supply
Dosage & Administration
- Usual dose: 5 mg once daily
- Maximum dose: 10 mg once daily
- May be taken without regard to meals
Interaction
- Potentially hazardous interactions with p-blockers
- No interaction with Digoxin
- No interaction with Cimetidine
- No significant alteration with Warfarin
- Food does not alter absorption
Contraindications
- Hypersensitivity to dihydropyridine derivatives
- Pregnant women

Pregnancy & Lactation
- Pregnancy Category C
- Only if potential benefit justifies potential risk
- Recommendation to discontinue nursing while Amlodipine is administered
Precautions & Warnings
- Precaution in patients with hepatic impairment
- Precaution during pregnancy and breastfeeding
Use in Special Populations
- Child dosing: 2.5 mg once daily for 6-17 years
- Elderly tolerance
- Normal dosage for renal impairment
- Caution in hepatic impairment: start at 2.5 mg once daily
Overdose Effects
- Excessive peripheral vasodilatation
- Reflex tachycardia
- Marked and prolonged hypotension
- Shock with fatal outcome
- Active cardiovascular support needed
- Vasoconstrictor and calcium gluconate may be beneficial
- Gastric lavage and charcoal use may be worthwhile
- Dialysis unlikely to be beneficial
