Therapeutic Class

  • First generation Cephalosporins

Indications

  • Treatment of infections caused by sensitive Gram-positive and Gram-negative bacteria
  • Upper respiratory tract infections: sinusitis, pharyngitis, tonsillitis, laryngo-tracheobronchitis, otitis media
  • Lower respiratory tract infections: bronchitis, lobar pneumonia, bronchopneumonia
  • Urinary tract infections: cystitis, urethritis, pyelonephritis
  • Skin and soft tissue infections: abscess, cellulitis, furunculosis, impetigo

Pharmacology

  • Semisynthetic broad spectrum bactericidal antibiotic
  • Active against both gram-positive and gram-negative microorganisms
  • Inhibits cross-linking process in bacteria cell walls

Dosage & Administration

  • Different doses for adults and children for various infections
  • Maximum daily dosage: 4 gm for adults
  • Dosage adjustment necessary in renal impairment
  • Injectable administration for adults and children
  • Prophylactic preoperative dose for adults

Interaction

  • Nephrotoxic drugs may increase risk of kidney damage
  • Diuretics and probenecid enhance possibility of renal toxicity

  • Hypersensitivity to cephalosporins

Precautions & Warnings

  • Positive result in Coombs’ testing
  • May cause false positive urine glucose result
  • Lactose content caution for patients with specific deficiencies

Use in Special Populations

  • Recommended doses for patients with renal impairment
  • Pediatric dosages should not exceed those specified for adults
  • Higher doses may be required for chronic or severe infections
  • Regular monitoring for urinary tract infections
  • Transfer from intramuscular/intravenous to oral treatment

Overdose Effects

  • Non-specific symptoms: nausea, vomiting, diarrhea, gastric upsets

  • Gastric lavage if large amount ingested

Storage Conditions

  • Freshly prepare G-Cefradine Suspension
  • Reconstituted Suspension should be used within 7-14 days
  • Injection solutions should be used within 2 hours at room temperature
  • Retain potency for 12 hours at 5°C
