Description
- Polycef is a first generation Cephalosporin indicated for the treatment of infections caused by sensitive Gram-positive and Gram-negative bacteria.
Indications
- Treatment of upper respiratory tract infections like sinusitis, pharyngitis, tonsillitis, laryngo-tracheo bronchitis, and otitis media
- Treatment of lower respiratory tract infections like bronchitis, lobar pneumonia, and bronchopneumonia
- Treatment of urinary tract infections like cystitis, urethritis, and pyelonephritis
- Treatment of skin and soft tissue infections like abscess, cellulitis, furunculosis, and impetigo
Pharmacology
- Cephradine is a semisynthetic broad spectrum bactericidal antibiotic active against both gram-positive and gram-negative microorganisms.
Dosage
- Dosage varies for different types of infections and age groups, ranging from 250mg to 4gm daily for adults and 25 to 100mg/kg for children.
- Dosage adjustment is necessary in patients with renal impairment.
- For injectable administration, the usual dose is 2-4 gm daily in four equally divided doses up to 8 gm daily for adults.
Interaction
- Concomitant use of nephrotoxic drugs such as aminoglycosides with Cefradine may increase the risk of kidney damage.
- Diuretics and probenecid enhance the possibility of renal toxicity.
Contraindications
- Cephradine should not be used in patients with known or suspected hypersensitivity to cephalosporins.
Side Effects
- May include gastro-intestinal disturbances, hypersensitivity phenomena, skin reactions, blood disorders, immune system disorders, psychiatric disorders, nervous system disorders, and renal and urinary disorders.
Pregnancy & Lactation
- Safety in pregnancy has not been established and should be used with caution in lactating mothers.
Precautions & Warnings
- Prolonged use of Polycef may result in superinfection due to the emergence of resistant organisms.
- Polycef should be administered with care to patients hypersensitive to penicillins because of the risk of cross-sensitivity between beta-lactam antibiotics.
- Polycef may cause a false positive urine glucose result when using certain testing solutions.
Use in Special Populations
- Recommendations for patients with renal impairment and those on chronic, intermittent haemodialysis.
Overdose Effects
- Symptoms of overdose are non-specific and may include nausea, vomiting, and diarrhoea. Supportive treatment is mainly advised.

Storage Conditions
- Polycef Suspension should be freshly prepared and used within a certain time frame if kept at room temperature or in a refrigerator.
- Polycef Injection solutions should be used within specified timeframes and must not be used beyond the date of expiry.
