Indications

  • Essential hypertension

Pharmacology

  • Dihydropyridine calcium-channel blocker
  • Long duration of action
  • Vasodilation
  • Reduced peripheral resistance
  • Increased coronary blood flow
  • Reflex increase in coronary heart rate

Dosage & Administration

  • Usual dose is 5 mg once daily
  • Maximum dose is 10 mg once daily
  • May be taken without regard to meals

Interaction

  • Potentially hazardous interactions with p-blockers in impaired cardiac left ventricular function
  • No interaction between Nelod and Digoxin in healthy volunteers
  • No interaction between Nelod and Cimetidine in healthy volunteers
  • Nelod did not significantly alter the effect of Warfarin on prothrombin time
  • Food does not alter the rate or extent of absorption of Nelod

Contraindications

  • Hypersensitivity to dihydropyridine derivatives
  • Pregnant women

Pregnancy & Lactation

  • Pregnancy Category C
  • Amlodipine should be used during pregnancy only if the potential benefit justifies the potential risk to the fetus
  • Nursing should be discontinued while Amlodipine is administered

Precautions & Warnings

  • Precaution in patients with hepatic impairment and during pregnancy and breastfeeding

Use in Special Populations

  • Dosage for children with hypertension from 6 to 17 years
  • Effect of amlodipine on blood pressure in patients less than 6 years of age is not known
  • Nelod used at similar doses in elderly or younger patients is equally well tolerated
  • Normal dosage regimens recommended in the elderly
  • Recommended dosage in renal impairment
  • Dosage recommendations in patients with mild to moderate hepatic impairment
  • Nelod should be initiated at the lowest dose and titrated slowly in severe hepatic impairment

Overdose Effects

  • Symptoms of large overdosage
  • Management of clinically significant hypotension due to amlodipine overdosage
