Dosage
- Initial antihypertensive oral dose of Amlodipine: 5 mg once daily
- Initial Amlodipine dose for chronic stable or vasospastic angina: 5-10 mg
- Recommended starting dose of Atorvastatin: 10 mg daily
- Dosage range for Atorvastatin: 10 to 80 mg once daily
- Atorvastatin administration: can be administered as a single dose at any time of the day with or without food
Mode of Action
- Amlodipine: peripheral arterial vasodilator
- Amlodipine: reduction in peripheral vascular resistance and blood pressure
- Atorvastatin: synthetic lipid-lowering agent
- Atorvastatin: HMG-CoA reductase inhibitor
Interaction
- Drug interaction with Amlodipine: potentiation of other antihypertensive drugs' effects
- Amlodipine did not affect the pharmacokinetics of Atorvastatin, Digoxin, Warfarin, or Cyclosporine
Contraindications
- Amlodipine: known hypersensitivity to Amlodipine
- Atorvastatin: hypersensitivity to any component of this medication
- Atorvastatin: active liver disease or unexplained persistent elevations of serum transaminases exceeding three times the upper limit of normal
Side Effects
- Amlodipine-induced vasodilatation gradual in onset, caution advised for acute hypotension
- Atorvastatin may cause elevation in serum creatine phosphokinase levels
- Commonly reported Atorvastatin adverse effects: constipation, flatulence, dyspepsia, headache, nausea, myalgia, diarrhea, asthenia, insomnia
Pregnancy & Lactation
- Safety not established in pregnancy
- Use of HMG-CoA reductase inhibitors during breastfeeding not recommended
Precautions & Warnings
- Caution advised with Amlodipine in patients with severe aortic stenosis
- Amlodipine use in patients with congestive heart failure to be exercised with caution
- Atorvastatin therapy should be discontinued if markedly elevated CPK levels occur
Pediatric Uses
- Safety and efficacy of Atorvastatin not established in children

Indications
- Treatment of hypertension
- Treatment of chronic stable angina
- Adjunct to diet for hypercholesterolemia
- Reduction of risk of nonfatal MI and nonfatal stroke in hypertensive patients with multiple risk factors for CHD
Pharmacology
- Amlodipine: calcium channel blocker
- Atorvastatin: statin (HMG-CoA reductase inhibitor)
- Amlodipine acts directly on vascular smooth muscle to reduce peripheral vascular resistance
- Atorvastatin inhibits the conversion of HMG-CoA to mevatonate, a rate limiting step in the synthesis of cholesterol
