Indications
- Treatment of upper respiratory tract infections (URTI)
- Treatment of lower respiratory tract infections
- Treatment of urinary tract infections (UTI)
- Bactericidal activity against Gram-positive organisms
- Bactericidal activity against Gram-negative organisms
Pharmacology
- Third generation semisynthetic cephalosporin antibiotic
- Bactericidal to both gram positive and gram negative bacteria
- Interference in the synthesis of bacterial cell wall
- High stability in the presence of Beta-lactamase enzyme
- About 40% to 50% absorption whether administered with or without food
Dosage & Administration
- Usual course of treatment is 7 days
- Recommended adults dosage is 200-400 mg daily
- Recommended children dosage is 8 mg/kg/day
- Recommended dosage for children in typhoid is 5 mg/kg body weight twice daily for 10-14 days
- Dosage in renal impairment
Interaction
- Increases in prothrombin times noted in some patients
Contraindications
- Known hypersensitivity to cephalosporin antibiotics
Side Effects
- Diarrhoea and stool changes
- Nausea, abdominal pain, dyspepsia, vomiting, flatulence
- Pseudomembraneous colitis
- Headache and dizziness
- Allergies including rash, pruritis, urticaria, drug fever, arthralgia
- Thrombocytopenia, leukopenia, eosinophilia
- Mild transient change in liver and renal function tests
- Genital pruritis and vaginitis
Pregnancy & Lactation
- No adequate and well-controlled studies in pregnant women
- Caution advised when administered to a nursing woman
Precautions & Warnings
- Caution advised for patients with hypersensitivity to other drugs
- Caution advised for penicillin-sensitive patients
- Caution advised for patients with markedly impaired renal function
Overdose Effects
- Gastric Lavage may be indicated
- No specific antidote exists
- Afixime not removed in significant quantities by dialysis
- Adverse reactions observed in healthy adult volunteers receiving single doses up to 2 g
