Pharmacology

Ceftriaxone is a 3rd generation broad-spectrum parenteral cephalosporin antibiotic with potent bactericidal activity against a wide range of Gram-positive and Gram-negative organisms.

Dosage

  • 1 to 2 gm for adults by intravenous or intramuscular administration once a day
  • 250 mg for uncomplicated gonococcal infections

Administration

  • Preparation for Intramuscular / Intravenous Injections
  • Injection administered over 2-4 minutes
  • Patient tolerance test recommended before administration

Contraindications

Not for patients with a history of hypersensitivity to cephalosporin antibiotics

Side Effects

  • Gastro-intestinal effects including diarrhea, nausea and vomiting
  • Cutaneous reactions including rash, pruritus, urticaria
  • Hematologic reactions including eosinophilia, thrombocytopenia
  • Hepatic reactions including elevations of SGOT or SGPT
  • CNS reactions including nervousness, confusion, sleep disturbances
  • Local phlebitis following intravenous administration

Pregnancy & Lactation

Not established for use in pregnancy, caution advised during lactation

Precautions & Warnings

  • Risk of anaphylactic shock
  • Possibility of gallbladder sludge
  • Regular blood picture checks during prolonged treatment

Use in Special Populations

Not for neonates if premature and newborn (up to 28 days of age)

Overdose Effects

No specific antidote, treatment should be symptomatic
