Indications

Pharmacology
Ceftriaxone is a 3rd generation broad-spectrum parenteral cephalosporin antibiotic with potent bactericidal activity against a wide range of Gram-positive and Gram-negative organisms. It kills bacteria by interfering with the synthesis of the bacterial cell wall. Ceftriaxone has a high degree of stability in the presence of beta lactamases and a relatively long plasma elimination half-life of about 6 to 9 hours.
Dosage
- 1 to 2 gm by intravenous or intramuscular administration once a day
- 250 mg IM for uncomplicated gonococcal infections
- Doses for infants and children
- Duration of therapy: 4 to 14 days

Contraindications
Should not be given to patients with a history of hypersensitivity to cephalosporin antibiotics
Side Effects
- Gastro-intestinal effects including diarrhea, nausea, and vomiting
- Cutaneous reactions including rash, pruritus, and urticaria
- Hematologic reactions including eosinophilia, thrombocytopenia, and leucopenia
- Hepatic reactions including elevations of SGOT or SGPT
- CNS reactions including nervousness, sleep disturbances, and headache
- Rare cases of local phlebitis following intravenous administration
Pregnancy & Lactation
Safety in human pregnancy not established. Low concentrations excreted in human milk.
Precautions & Warnings
- Anaphylactic shock precautions
- Gallbladder sludge findings in rare cases
- Regular blood picture check during prolonged treatment
Use in Special Populations
Must not be given to neonates if the neonate is premature and newborn (up to 28 days of age)
Overdose Effects
No specific antidote. Treatment should be symptomatic.
