Indications

Pharmacology

  • Imipenem - penem antibacterial drug
  • Cilastatin - renal dehydropeptidase inhibitor
  • Bactericidal activity from inhibition of cell wall synthesis
  • Stability in the presence of beta-lactamases
  • Inhibition of penicillin binding proteins in bacteria

Dosage & Administration

  • Dosage based on suspected or confirmed pathogen susceptibility
  • Different dosage regimens for adult patients with normal renal function
  • Reduction in dose required for patients with reduced renal function
  • Reconstitution required with appropriate diluent
  • Intravenous infusion administration

Interaction

  • Concurrent admin with probenecid may increase cilastatin half-life
  • Increased risk of seizures when used with ganciclovir

Contraindications

  • Hypersensitivity to any component of the combination

  • Nausea, diarrhea, vomiting
  • Rash, pruritus, urticaria
  • Pain at injection site, erythema
  • Fever
  • Hypotension
  • Seizures, dizziness, somnolence

Pregnancy & Lactation

  • Pregnancy Category C
  • Caution advised during lactation
  • No adequate and well-controlled studies in pregnant women

Precautions & Warnings

  • Serious and occasionally fatal hypersensitivity reactions reported
  • Seizure potential with CNS adverse reactions

Reconstitution

  • Do not use diluents containing benzyl alcohol for neonates
  • Reconstitution with 0.9% Sodium Chloride Injection
  • Reconstituted solutions range from colorless to yellow
  • Shake vial well before transferring to infusion solution
  • Visual inspection for particulate matter and discoloration required

Storage Conditions

  • Store at or below 25°C temperature
  • Keep away from light and wet place
  • Keep out of reach of children
