Pharmacology
- Main mechanism of action on protein synthesis
- Energy-dependent active transport system
- Binds to the 30s ribosomes
- Inhibit bacterial protein synthesis
- Susceptibility to various bacteria
- Urinary concentrations for E. coli
- Active against anaerobic species of bacteria
- High concentrations in the gastrointestinal contents
- Half-life of about 12 hours
- Excreted in the urine and in the faeces
Dosage & Administration
- Usual adult oral dosage of 1-2g daily
- Usual oral dosage for children
- Dosage for specific indications like Acne vulgaris, staphylococcal infections, streptococcal infections, amoebiasis, brucellosis, subacute bacterial endocarditis, syphilis
Interaction
- Impaired absorption with antacids containing certain cations
- Interference with the bactericidal action of penicillin
- Potentiation of the effect of anticoagulants
- Decrease efficacy of oral contraceptives
- Potential increase in hypoglycaemic effect of insulin and sulfonylureas
- Potential increase in toxic effects of ergot alkaloids and methotrexate
Contraindications
- Hypersensitivity to tetracycline groups
- Cross-sensitivity with other tetracycline members
- Systemic lupus erythematosus
- Renal impairment
