Pharmacology
- Mechanism of action on protein synthesis
- Susceptibility of Gram positive aerobic Cocci
- Resistance of staphylococci and streptococci

- Activity against anaerobic species of bacteria
- Absorption from the gastro-intestinal tract
- Distribution through body tissues and fluids
- Excretion in the urine and faeces
- Half-life of 12 hours
Dosage & Administration
- Usual adult oral dosage of 1-2g daily
- Usual oral dosage for children older than 8 years
- Administration preferably 1 hour before or 2 hours after meals
- Specific indications and recommended dosages for different infections
Interaction
- Impaired absorption with antacids containing certain cations
- Interference with bactericidal action of penicillin
- Potentiation of anticoagulants
- Decrease in efficacy of oral contraceptives
- Exacerbation of nephrotoxic effects by diuretics
- Increase in hypoglycaemic effect of insulin and sulfonylureas in patients with DM
- Increase in toxic effects of ergot alkaloids and methotrexate
Contraindications
- Hypersensitivity to tetracycline groups
- Cross-sensitivity in patients with systemic lupus erythematosus
- Renal impairment

- Depression of bone growth
- Gray-brown discoloration of the teeth
- Enamel hypoplasia
- Hypersensitivity reactions
- Photosensitivity reactions
- Gastrointestinal effects
- Accentuated prerenal azotemia
- Superinfections with oral and anogenital candidiasis
- Esophageal ulcerations
Pregnancy & Lactation
- Risk of hypertoxicity in the mother
- Effects on the developing fetus
- Impaired bone growth and permanent discoloration of the child's teeth
Precautions & Warnings
- Care with impaired liver function
- Avoidance of high doses
- Avoidance of concomitant use with potentially hepatotoxic drugs
