Pharmacology
- Mechanism of action on protein synthesis
- Active transport system
- Susceptibility to Gram positive aerobic Cocci
- Resistance in staphylococci and streptococci
- Resistance in Pseudomonas and many Enterobacteriaceae
- Activity against anaerobic species of bacteria
- Distribution in body tissues and fluids
- Absorption from the gastrointestinal tract
- Excretion in urine and faeces
- Half-life of about 12 hours
Dosage & Administration
- Usual adult oral dosage of 1-2 g daily
- Usual oral dosage for children older than 8 years
- Administration preferably one hour before or 2 hours after meals
- Specific indications and dosage for acne vulgaris, staphyococcal infections, streptococcal infections, amoebiasis, brucellosis
- Duration of therapy determined by individual progress
Interaction
- Impaired absorption with antacids containing divalent and trivalent cations
- Interference with bactericidal action of penicillin
- Potentiation of anticoagulants
- Decrease efficacy of oral contraceptives
- Nephrotoxic effects exacerbated by diuretics or other nephrotoxic drugs
- Increase in hypoglycaemic effect of insulin and sulfonylureas
- Increase in toxic effects of ergot alkaloids and methotrexate
Contraindications
- Hypersensitivity to tetracycline groups
- Systemic lupus erythematosus
- Renal impairment
Side Effects
- Depression of bone growth
- Permanent gray-brown discoloration of the teeth
- Enamel hypoplasia
- Hypersensitivity reactions
- Photosensitivity reactions
- Gastrointestinal effects
- Accentuated prerenal azotemia
- Superinfections with oral and anogenital candidiasis
- Esophageal ulcerations
Pregnancy & Lactation
- Not recommended during pregnancy
- Risk of hypertoxicity in the mother and effects on the developing fetus
- Potential impaired bone growth and permanent discoloration in children during breastfeeding and up to a certain age
Precautions & Warnings
- Care for patients with impaired liver function
- Avoid high doses
- Avoid concomitant use with potentially hepatotoxic drugs
