Description

  • Primarily a bacteriostatic broad-spectrum antibiotic
  • Wide spectrum of activity against Gram-positive and Gram-negative microorganisms
  • Acts as an antimicrobial agent by inhibiting bacterial protein synthesis
  • Effective against parasites such as rickettsia, mycoplasma, chlamydia and amoeba

Pharmacology

  • Mechanism of action: Inhibits cell growth by inhibiting translation
  • Binds to the 30S ribosomal subunit
  • Prevents the amino-acyl tRNA from binding to the A site of the ribosome
  • Lipophilic and can easily pass through the cell membrane or passively diffuses through porin channels in the bacterial membrane

Dosage & Administration

  • Adults: 250-500 mg every 6 hours
  • Tetracyclines should be given one hour before or two hours after meals to avoid interference with food, milk, and some dairy products

Interactions

  • Antacids containing Aluminium, Calcium, Magnesium, Zinc, or Iron salts may impair the absorption of Oramycin
  • Long term therapy may depress plasma prothrombin activity and require reduced doses of concomitant anticoagulants
  • Concurrent use with oral contraceptives may lead to pregnancy or breakthrough bleeding

Contraindications

  • Hypersensitivity to Tetracyclines
  • Children under 12 years of age
  • Pregnancy due to possible tooth staining in the fetus and growth retardation effects
