Description
- Primarily a bacteriostatic broad-spectrum antibiotic
- Wide spectrum of activity against Gram-positive and Gram-negative microorganisms
- Acts as an antimicrobial agent by inhibiting bacterial protein synthesis
- Effective against parasites such as rickettsia, mycoplasma, chlamydia and amoeba
Indications
- Respiratory tract infections
- Mycoplasma pneumonia
- Urinary tract infections
- Infections due to Chlamydia trachomatis
- Uncomplicated urethral, endocervical or rectal infections
- Non-gonococcal urethritis
- Chancroid
- Granuloma inguinale
- Lymphogranuloma venereum
- Gonorrhea
- Syphilis
- Skin infections: Acne and rosacea
- Ophthalmic infections: Trachoma
- Rickettsial infections
- Other infections: Psittacosis, brucellosis, cholera, acute intestinal amoebiasis
Pharmacology
- Mechanism of action: Inhibits cell growth by inhibiting translation
- Binds to the 30S ribosomal subunit
- Prevents the amino-acyl tRNA from binding to the A site of the ribosome
- Lipophilic and can easily pass through the cell membrane or passively diffuses through porin channels in the bacterial membrane
Dosage & Administration
- Adults: 250-500 mg every 6 hours
- Tetracyclines should be given one hour before or two hours after meals to avoid interference with food, milk, and some dairy products
Interactions
- Antacids containing Aluminium, Calcium, Magnesium, Zinc, or Iron salts may impair the absorption of Oramycin
- Long term therapy may depress plasma prothrombin activity and require reduced doses of concomitant anticoagulants
- Concurrent use with oral contraceptives may lead to pregnancy or breakthrough bleeding
Contraindications
- Hypersensitivity to Tetracyclines
- Children under 12 years of age
- Pregnancy due to possible tooth staining in the fetus and growth retardation effects

Pregnancy & Lactation
- Crosses the placenta and can have toxic effects on the developing fetus
- Embryotoxicity noted in animals treated early in pregnancy
