Pharmacology

  • Bacteriostatic antibiotic
  • Gram-negative strains sensitivity
  • Anaerobes sensitivity
  • Activity against Haemophilus influenzae

  • Active against Plasmodium falciparum

Dosage

  • 200mg on first day followed by 100mg daily for 7-10 days
  • Severe infections: 200mg daily for 10 days
  • Acne: 100mg daily
  • Uncomplicated genital chlamydia: 100mg twice daily for 7-21 days

Administration

Capsules should be swallowed whole with plenty of fluid during meals while sitting or standing.

Interaction

  • Impaired by antacids
  • Impaired by iron-containing preparations
  • Decreased half-life with barbiturates, carbamazepine, phenytoin
  • Render oral contraceptives less effective
  • May require downward adjustment of anticoagulant dosages
  • Avoid giving in conjunction with penicillin

Contraindications

  • Hypersensitivity to tetracyclines
  • Children under 8 years of age
  • Pregnant women
  • Lactating mothers

  • Nausea

  • Hemolytic anaemia
  • Eosinophilia

Pregnancy & Lactation

Avoid in pregnant women and lactating mothers due to risk of staining and effect on bone growth in the foetus

Avoid use during tooth development in children to prevent permanent discoloration of teeth
