Description
- Belongs to macrolides group of antibiotics
- Exerts bacteriostatic or bactericidal action based on interference with ribosomal protein synthesis within bacterial cell
- Effective against penicillinase producing staphylococci
- Susceptible to gram positive and gram negative micro-organisms
Pharmacology
- Inhibits microsomal protein synthesis in susceptible organisms by inhibiting the translocation process
- Active against many gram positive bacteria, some gram negative bacteria, mycoplasmas, and chlamydia
- Destroyed by acid, but stable ester form is administered
- Widely distributed throughout body tissue and fluids
Dosage & Administration
- Adult dose: 250-500 mg every six hours for mild to moderate infections, may increase in severe cases
- Elderly dose: No special recommendation, administered three times daily or twice daily
- Children aged 2 to 8 years: 30-50 mg/kg body weight per day divided into four equal dosages
- Infants and children upto 2 years: 30-50 mg/kg body weight in divided doses
Interaction
- Potential increase of serum theophylline levels and theophylline toxicity
- May need to reduce theophylline dose when using Mitrocin
Contraindications
- Known hypersensitivity to Erythromycin
Side Effects
- Rare allergic reactions, mild anaphylaxis
- Occasional abdominal discomfort, nausea, and vomiting after oral administration
Pregnancy & Lactation
- No evidence of teratogenicity or toxicity in humans
- Caution should be exercised during pregnancy and lactation as it crosses the placental barrier and excreted in breast milk
Precautions & Warnings
- Given with care in patients with impaired hepatic function as it is excreted principally in the bile
Overdose Effects
- Discontinue in case of overdosage
- Prompt elimination of unabsorbed drug is recommended
- Not removed by peritoneal dialysis or haemodialysis

Reconstitution
- Directions for reconstitution of suspension
- Shake the bottle to loosen powder
- Add boiled and cooled water to the dry powder of the bottle
- Shake well after each addition until all the powder is in suspension
- Reconstituted suspension should be stored in a cool and dry place, preferably in refrigerator

- Keep below 25°C temperature, away from light and moisture
- Keep out of the reach of children
