Indications
- Lower respiratory tract infections
- Upper respiratory tract infections
- Otitis media
- Skin and soft tissue infections
- Sexually transmitted diseases in men and women
Pharmacology
- Absorbed orally
- Peak concentration in adults: 2.1 to 3.2 hours
- High concentration in phagocytes
- Bacteriostatic levels in infected tissue for several days
- Active against aerobic and facultative gram-positive/negative microorganisms
Dosage
- Adult: 500 mg once daily orally for 3 days
- Children: 10 mg/kg body weight once daily for 3 days
- Azithromycin Injection for IV Infusion: 500 mg as a single daily dose
Reconstitution procedure
- Shake the bottle well to loosen the powder
- Add boiled and cooled water up to the water mark of the bottle label
- Shake until powder is completely mixed with water
Interaction
- Antacid: Take at least 1 hour before or 2 hours after
- Carbamazepine: No significant effect on plasma levels
- Cyclosporin: Caution before co-administration
- Digoxin: Monitor digoxin levels
- Ergot derivatives: Avoid co-administration
- Methylprednisolone: No significant effect
- Theophylline: No evidence of pharmacokinetic interaction
- Warfarin: Did not alter the anticoagulant effect
- Terfenadine: Did not affect the pharmacokinetics
Contraindications
- Hypersensitive to Azithromycin or macrolide antibiotic
- Co-administration of ergot derivatives
- Patients with hepatic diseases
Side Effects
- Gastrointestinal side-effects
- Allergic reactions such as rash or photosensitivity
- Rare reports of serious hypersensitivity reactions
- Elevations in liver transaminases
- Cholestatic jaundice
- Transient mild reductions in neutrophil counts
- Reversible hearing impairment
Pregnancy & Lactation
- Pregnancy Category B
- No evidence of harm to the fetus in animal reproduction studies
- Use during pregnancy if adequate alternatives are not available
- Caution when administered to nursing women
Precautions & Warnings
- Rare serious allergic reactions reported
- Caution in patients with renal impairment
- Not recommended in patients with hepatic disease
- No evidence of effect on ability to drive or use machines
Overdose Effects
- Symptoms may include hearing loss, severe nausea, vomiting, and diarrhea
- General supportive measures are indicated
