Macrolide with antibacterial properties similar to erythromycin.

Pharmacology

Inhibits protein synthesis by irreversibly binding to the 50s ribosomal subunits

Dosage & Administration

  • Usual Adult Dose: 300 mg tablet once daily or 150 mg tablet twice a day for at least 2 days
  • Usual Pediatric Dose: 2.5 to 5 mg/kg body weight twice a day

Absorption

Best administered on an empty stomach

Interaction

  • Cytochrome P-450 Affinity: None
  • Mild Interaction with: Theophylline
  • No Effect on:
    • Carbamazepine
    • Warfarin
    • Antacids
    • H2-receptor antagonists
    • Food

Contraindications

Simultaneous administration with ergotamine or its derivatives

Pregnancy & Lactation Safety

  • Safety not established in pregnancy
  • Appears in breast milk in small amounts with no adverse effects in breastfed infants

Precautions & Warnings

  • Not to be used in patients with a history of hypersensitivity to the drug
  • In patients with hepatic diseases, the dose should not exceed 150 mg twice a day
