Description

Linzolid is indicated for the treatment of nosocomial pneumonia, community-acquired pneumonia, complicated skin and skin structure infection, uncomplicated skin and skin structure infections, and Vancomycin-resistant Enterococcus faecium infections. It is a synthetic, antibacterial agent belonging to oxazolidinones with in vitro activity against Gram-positive and certain Gram-negative bacteria. Linezolid tablets should be taken in oral route with or without food. Linezolid IV infusion should be administered by intravenous infusion over a period of 30 to 120 minutes.

Dosage and Administration

  • Infections:
    • Nosocomial pneumonia
    • Community-acquired pneumonia
    • Complicated skin and skin structure infections
    • Uncomplicated skin and skin structure infections
    • Vancomycin-resistant Enterococcus faecium infections
  • Recommended Dosage:
    • Pediatric Patients: 10 mg/kg orally or intravenously every 8 hours
    • Adults and Adolescent: 600 mg orally or intravenously every 12 hours
    • Recommended Duration of Treatment: 10 to 28 consecutive days
  • Intravenous Administration: Administered by intravenous infusion over a period of 30 to 120 minutes
  • Hepatic Impairment: No dose adjustment is recommended for patients with mild-to-moderate hepatic impairment
  • Use in Children and Adolescents: The dose of linezolid for children and adolescents have been described

Interaction

  • Drug Interaction with Medication: Linzolid is a reversible, nonselective inhibitor of monoamine oxidase
  • Interaction with Food and Others: Advice patients to avoid large quantities of foods or beverages with high tyramine content while taking Linzolid

Contraindications

  • Known hypersensitivity to Linezolid or any of the other product components
  • Patients taking any medicinal product which inhibits monoamine oxidases A or B
  • Uncontrolled hypertension, pheochromocytoma, thyrotoxicosis, carcinoid syndrome
  • Patients taking directly and indirectly acting sympathomimetic agents, vasopressive agents, dopaminergic agents, serotonin re-uptake inhibitors, tricyclic antidepressants, serotonin 5-HT1 receptor agonists, meperidine or buspirone

Side Effects

    • Hypertension
    • Dyspepsia
    • Localized abdominal pain
    • Pruritus

Pregnancy and Lactation

  • Pregnancy: Available data with linezolid use in pregnant women have not identified a drug-associated risk of major birth defects, miscarriage or adverse maternal or fetal outcomes
  • Lactation: Linezolid is present in breast milk. Advise lactating women to monitor a breastfed infant for diarrhea and vomiting

Precautions and Warnings

  • Complete blood counts should be monitored weekly in patients who receive Linzolid
  • Prompt ophthalmic evaluation is recommended if patients experience symptoms of visual impairment
  • Cautious use in patients with catheter-related bloodstream infections or catheter-site infections
  • Careful monitoring for potential increases in blood pressure
  • Immediate medical evaluation for patients with recurrent nausea or vomiting, unexplained acidosis or low bicarbonate level
  • Dose adjustment may be required if hypoglycemia occurs

Overdose Effects

No cases of overdose have been reported. Symptomatic and supportive care is advised together with maintenance of glomerular filtration
