Indications

• Upper respiratory tract infections
• Lower respiratory tract infections
• Genito-urinary tract infections
• Skin and soft tissue infections
• Bone and joint infections
• Other infections

Composition

• 375 mg tablet
• 625 mg tablet
• 1 gm tablet
• Powder for Suspension
• Powder for Suspension (Forte)
• 1.2 gm Injection
• 0.6 gm injection

Pharmacology

• Antibacterial combination
• Broad spectrum bactericidal activity
• Spectrum of activity against Gram-positive & Gram-negative microorganisms
• Resistance to degradation by beta-lactamases
• Inactivation of beta-lactamase enzymes

Dosage

• Tablet for adults and children over 12 years
• Suspension for children of different age groups
• Forte suspension for recommended daily dosage
• IV Injection for adults and children
• Increased dosage for more serious infections

Administration

• Oral dosage form without regard to meals
• Enhanced absorption at the start of a meal
• IV injection suitable for intravenous administration
• Requirement to use reconstituted vial within 20 minutes

Interaction

• Prolongation of bleeding time and prothrombin time
• Reduction in efficacy of oral contraceptives
• Concomitant use of allopurinol
• Potential cross-sensitivity with other beta-lactam antibiotics

Contraindications

• Penicillin hypersensitivity
• Cross-sensitivity with other beta-lactam antibiotics
• History of Co-amoxiclav or Penicillin-associated cholestatic jaundice

Side Effects

• Rare and transitory nature of side effects
• Gastrointestinal side effects
• Hepatitis and cholestatic jaundice
• Allergic skin reactions
• Angioedema and anaphylaxis

Pregnancy & Lactation

• Animal studies on teratogenic effect
• Limited use in human pregnancy without untoward effect
• Detection of Amoxicillin in breast milk

Precautions & Warnings

• Careful use in patients on anticoagulation therapy
• Dosage adjustment in patients with renal impairment
• Maintenance of adequate fluid intake and urinary output

Use in Special Populations

• Dose adjustment in patients with renal impairment
• Administration hepatic impairment
• Reduction in dosage for children

Overdose Effects

• Unlikelihood of problems with overdose
• Possible gastrointestinal symptoms
• Potential removal by haemodialysis

Reconstitution

• Reconstitution of IV injection
• Incompatibility with certain solutions for injection
• Injection into infusion fluids containing glucose, bicarbonate and dextran

• Limited use within 7 days of reconstitution
