DosageAndAdministration

  • Concentrations of no more than 5 mg/ml
  • Rates of no more than 10 mg/min
  • Usual daily dose for adults: 2 gm (in 4 or 2 divided doses)
  • Total daily dose for children: 40 mg/Kg (in 4 divided doses)
  • Initial dose for infants and neonates: 15 mg/Kg
  • Dosage adjustment for impaired renal function
  • Capsule dosage for adults with antibiotic-associated pseudomembranous colitis: 125 to 500 mg administered orally every 6 to 8 hours for 7 to 10 days
  • Capsule dosage for pediatrics: approximately 40 mg/kg in 3 or 4 divided doses for 7 to 10 days administered orally

Pharmacology

  • Binding to D-alanyl-D-alanine portion cell wall precursor
  • Blockage of glycopeptide polymerisation
  • Immediate inhibition of cell wall synthesis
  • Secondary damage to the cytoplasmic membrane
