Description
- Glycopeptide-antibiotic for treating various infections
- Produced by Actinoplanes teichomyceticus
- Sterile, pyrogen-free ivory white powder for reconstitution with water for injection
- Freely soluble in water and gives a clear solution on reconstitution
- Inhibits cell-wall biosynthesis of susceptible organisms
- Interferes with peptidoglycan synthesis
- Can be given intravenously or intramuscularly
- Dosage should be adapted based on infection severity and patient's response
- Contraindicated in patients with known hypersensitivity to the drug
- May cause local (erythema, injection site abscess), allergic (rash, pruritus), gastrointestinal (nausea, vomiting), CNS (weakness, dizziness) and hearing (ringing in ears) reactions
- Pregnancy Category B3 - should be used with caution during pregnancy and lactation
- Should be administered with caution in patients known to be hypersensitive to Vancomycin

Indications
- Complicated skin and soft tissue infections
- Bone and joint infections
- Hospital-acquired pneumonia
- Community-acquired pneumonia
- Complicated urinary tract infections
- Infective endocarditis
- Alternative oral treatment for Clostridium difficile infection-associated diarrhoea and colitis
Mode of Action
- Interferes with cell-wall biosynthesis at a site different from beta-lactams
- Specific binding to D-alanyl-D-alanine residues
Dosage & Administration
- Given intravenously (bolus injection or infusion) or intramuscularly
- Dosage adjusted based on infection severity and patient's kidney function
- Appropriate for adults, elderly, and children with normal or kidney failure
- Recommended dosage for various infections and age groups
Interaction
- Administered with caution in patients receiving concurrent nephrotoxic or ototoxic drugs
Contraindications
- Contraindicated in patients with known Hypersensitivity to the drug
Side Effects
- Local reaction: erythema, injection site abscess when given intramuscularly
- Allergic reaction: rash, pruritus, fever, shivering, anaphylactic reactions
- Gastrointestinal reactions: nausea, vomiting, diarrhea
- CNS reactions: physical weakness, dizziness, headache
- Hearing reactions: ringing in the ears, balance disorders
Pregnancy & Lactation
- Pregnancy Category B3 - should not be used during pregnancy or lactation unless benefits outweigh risks

Reconstitution
- Inject slowly the water into the powder vial containing active ingredient
- Rotate the vial gently until all the powder is dissolved

- Store at a temperature not exceeding 30°C in a dry place
- Reconstituted solution should be used immediately or stored under refrigeration for 24 hours maximum
