Indications

• Bactericidal action against a wide range of organisms
• Treatment of respiratory tract infections
• Treatment of genito-urinary tract infections

• Treatment of septicaemia
• Adjuvant treatment for cholera

• Usable for chronic cystitis

Pharmacology

• Broad spectrum bactericidal activity
• Activity against gram-positive bacteria
• Activity against gram-negative bacteria
• Activity against some protozoa
• Exerts bactericidal action by sequential blockade of bacterial enzyme systems

Dosage & Administration

• Dosage for mild to moderate infections for adults
• Dosage for severe infections for adults
• Long term therapy dosage for adults
• Gonorrhoea treatment dosage for adults
• Dosage for mild to moderate infections for children
• Dosage for severe infections for children
• Dosage for long term therapy for children

Contraindications

• Hypersensitivity to trimethoprim or sulphonamides
• Megaloblastic anaemia due to folate deficiency
• Marked liver parenchymal damage
• Blood dyscrasia
• Severe renal insufficiency
• Glucose 6-phosphate dehydrogenase deficiency

Side Effects

• Crystalluria
• Allergic reactions
• Haemolysis
• Thrombocytopenia
• Neutropenia
• Agranulocytosis
• Malaise
• Headache
• Nausea
• Vomiting

Pregnancy & Lactation

• Sulphonamides pass the placenta
• Excreted in breast milk
• May cause kernicterus

Precautions & Warnings

• Risk of macrocytic anaemia with prolonged full dose treatment
• Interference with Folic acid conversion
• Possible potentiation of action of sulphonylurea in diabetic patients
