Indications

Pharmacology

  • Broad-spectrum bactericidal activity
  • Acts against gram-positive & gram-negative bacteria
  • Inhibits bacterial enzyme systems
  • Exerts action by blocking biosynthesis of folinic acid

Dosage & Administration

  • Double strength tablet for over 12 years
  • Tablet for over 12 years
  • Suspension for under 12 years
  • Different dosages for mild, moderate, and severe infections
  • Specific dosages for long-term therapy and gonorrhoea

Contraindications

  • Hypersensitivity to trimethoprim or sulphonamides
  • Megaloblastic anaemia due to folate deficiency
  • Liver parenchymal damage and blood dyscrasia
  • Severe renal insufficiency and glucose 6-phosphate dehydrogenase deficiency

Side Effects

  • Crystalluria
  • Allergic reactions
  • Haemolysis
  • Thrombocytopenia
  • Neutropenia
  • Agranulocytosis
  • Less serious effects like malaise, headache, nausea, and vomiting

Pregnancy & Lactation

  • Sulphonamides pass the placenta
  • Excreted in breast milk
  • May cause kernicterus

Precautions & Warnings

  • Risk of macrocytic anaemia
  • Careful with diabetic patients receiving sulphonylurea drug
