Description

  • Bactericidal in vitro to a wide range of Gram-positive and Gram-negative organisms

  • Exerts its bactericidal action by the sequential blockade of two bacterial enzyme systems in the biosynthesis of folinic acid in the microorganisms

Indications

  • Bactericidal activity against a wide range of organisms including Streptococcus, Staphylococcus, Pneumococcus, Neisseria, E. coli, Haemophilus, Salmonella, Shigella, Vibrio cholerae, Brucella, Pneumocystis carinii, Nocardia, and Bordetella
  • Suitable for respiratory, genito-urinary, gastro-intestinal, and other infections
  • Particularly suitable for the treatment of chronic bronchitis and urinary tract infections

DosageAndAdministration

Contraindications

  • Hypersensitivity to trimethoprim or sulphonamides
  • Megaloblastic anaemia due to folate deficiency
  • Liver parenchymal damage, blood dyscrasia, severe renal insufficiency, glucose 6-phosphate dehydrogenase deficiency

SideEffects

  • Rare side effects such as crystalluria, allergic reactions, haemolysis, thrombocytopenia, neutropenia, agranulocytosis
  • Less serious side effects include malaise, headache, nausea, and vomiting

PregnancyAndLactation

  • Excretion in breast milk may cause kernicterus

PrecautionsAndWarnings

  • Risk of macrocytic anaemia with prolonged full dose treatment
  • Possible potentiation of action of sulphonylurea in diabetic patients receiving sulphonylurea drug
