Description
Spardon is a broad-spectrum antibacterial agent used for the treatment of various infections caused by susceptible strains of designated microorganisms. It is also found to be effective in treating bacterial infections in the respiratory tract and lungs.

- Community-acquired pneumonia
- Acute bacterial exacerbations of chronic bronchitis
- Chronic Obstructive Pulmonary Disease
- Urinary tract infections
- Bacterial prostatitis
- Bacterial diarrhea
- Osteomyelitis
- Tuberculosis treatment
- Leprosy treatment
Mode of action
- Inhibition of DNA gyrase and topoisomerase IV
- Effective against gram positive organisms
- More effective than other fluoroquinolones against certain bacteria
Absorption
- Oral bioavailability of 92%
- Good absorption following oral administration
- Unaffected by administration with milk or food
Distribution
- Well-distributed into body fluids and tissues
- Low plasma protein binding at about 45%
- Concentrates well in lower respiratory tract tissues
- High concentration in alveolar macrophages
Metabolism
- Metabolized by the liver primarily by phase II glucuronidation
- Metabolism does not utilize or interfere with cytochrome-mediated oxidation
Excretion
- Excreted in both feces and urine
- Approximately 10% excreted in the urine as unchanged drug
Dosage and administration
- Recommended daily dose for patients with normal renal function
- Loading dose on the first day followed by maintenance dose
- Adjusted dosage for patients with renal impairment
Interaction
- Chelation complex formation with aluminium and magnesium cations in antacids and sucralfate
- Contraindicated with medications known to prolong the QTc interval
Contraindications
- Hypersensitivity to any of its ingredients
- Pregnancy and lactation
- Glucose-6-Phosphate Dehydrogenase (G6PD) deficiency
- History of Achilles tendinitis following the use of fluoroquinolones

- Mostly mild to moderate in severity
- Transient in nature
- Photosensitivity reaction
- Diarrhea
- Nausea
- Headache
- Dizziness
- Taste perversion
Pregnancy and lactation
- No adequate and well-controlled studies in pregnant women
- Use during pregnancy only if potential benefit justifies potential risk to the fetus
Precautions and warnings
- Discontinue therapy at first signs or symptoms of phototoxicity reaction
- Maintain adequate hydration to prevent highly concentrated urine
- Administer with caution in the presence of renal insufficiency
- Avoid excessive exposure to sunlight
Use in special populations
- Pharmacokinetics in geriatric, pediatric, and gender differences
- Impact of renal insufficiency on plasma concentrations
- No alteration in patients with mild or moderate hepatic impairment without cholestasis
