Indications

• Treatment of HIV-1 infection
• Treatment of chronic hepatitis B in adults

Dosage & Administration

• Recommended dose of 245 mg Tenofovir tablet
• Oral administration once daily
• No regard to food

Mode of Action

• Inhibits activity of HIV reverse transcriptase
• Competes with natural substrate deoxyadenosine 5-triphosphate

Pharmacokinetics

• Oral bioavailability approximately 25%
• Maximum serum concentrations achieved in 1.0-10.4 hrs
• Protein binding very low, <0.7% to human plasma proteins and <7.2% to serum proteins
• Half-life approximately 17 hours
• Eliminated by glomerular filtration and active tubular secretion

Interaction

• Cautious coadministration with didanosine
• Monitoring for adverse reactions with atazanavir and lopinavir/ritonavir

Contraindications

• Broken bone due to disease or illness
• Enlarged fatty liver
• Severe liver disease
• Acute kidney disease
• Low amount of phosphate in the blood
• Increased blood acidity due to high levels of lactic acid
• Overweight
• Breastfeeding
