Description
- Fungistatic medication for the treatment of various fungal infections
Indications
- Tinea capitis (ringworm of the scalp)
- Tinea corporis (ringworm of the body)
- Tinea pedis (athlete's foot)
- Tinea unguium (onychomycosis; ringworm of the nails)

Mode of Action
- Inhibits fungal cell mitosis and nuclear acid synthesis
- Binds to and interferes with the function of spindle and cytoplasmic microtubules
- Binds to keratin in human cells
Dosage
- Accurate diagnosis of infecting organism essential
- Medication must be continued until infecting organism is completely eradicated
- General measures in regard to hygiene should be observed
- Dosage varies for different types of infections and age groups
Administration
- Should be taken with food
- Take immediately after meals
Interaction
- May require dosage adjustment with warfarin-type anticoagulants
- Concomitant use of barbiturates may depress Griseofulvin M activity
- Reduces efficacy of oral contraceptives and may increase incidence of breakthrough bleeding
Contraindications
- Contraindicated in patients with porphyria, hepatocellular failure, and history of hypersensitivity to griseofulvin
- Should not be prescribed to pregnant patients
Side Effects
- Most commonly hypersensitivity reactions
- Paresthesias of the hands and feet
- Oral thrush, nausea, vomiting, and other gastrointestinal disturbances
- Rarely proteinuria and leukopenia
Pregnancy & Lactation
- Pregnancy Category C - potential benefit justifying the potential risk to the foetus
Precautions & Warnings
- Patients on prolonged therapy should be under close observation
- Periodic monitoring of organ system function should be done
- Possibility of cross sensitivity with penicillin exists
- Photosensitivity reaction occasionally associated with therapy
