Indications
- Tinea capitis (ringworm of the scalp)
- Tinea corporis (ringworm of the body)
- Tinea pedis (athlete's foot)
- Tinea unguium (onychomycosis; ringworm of the nails)
- Tinea cruris (ringworm of the thigh)
- Tinea barbae (barber's itch)
- Inhibits the growth of fungi such as Trichophyton rubrum, Trichophyton tonsurans, Trichophyton mentagrophytes, Microsporum canis, Epidermophyton floccosum, etc.
Pharmacology
- Fungistatic
- Inhibits fungal cell mitosis and nuclear acid synthesis
- Binds to and interferes with spindle and cytoplasmic microtubules
- Binds to keratin in human cells
Dosage
- Accurate diagnosis essential
- Medication duration depends on complete eradication of infecting organism
- Specific treatment periods for different infections
- Hygiene measures for controlling infection or reinfection
- Adult dosage: 500 mg daily for most cases, 1.0 gm for difficult cases
- Children dosage: 5 mg per pound of body weight per day
Administration
- Should be taken with food
- Take immediately after meals
Interaction
- Dosage adjustment of warfarin-type anticoagulant during and after Grisin therapy
- Concomitant use of barbiturates depresses Grisin activity
- Reduces the efficacy of oral contraceptives
- Increases the incidence of breakthrough bleeding
Contraindications
- Contraindicated in patients with porphyria, hepatocellular failure, and history of hypersensitivity to griseofulvin
- Not prescribed to pregnant patients due to reported cases of conjoined twins
Side Effects
- Most common adverse reactions are hypersensitivity type
- Rarely reported paresthesias, oral thrush, nausea, vomiting, etc.
- Rare reports of proteinuria and leukopenia
- Discontinue drug if granulocytopenia occurs

- Close observation for patients on prolonged therapy
- Periodic monitoring of organ system function
- Possibility of cross sensitivity with penicillin
- Warned against exposure to intense sunlight due to occasional photosensitivity reaction
- Aggravation of lupus erythematosus in case of photosensitivity reaction
