Pharmacology
- Potent inhibitor of fungal cytochrome P-450 dependent enzymes
- Responsible for the synthesis of ergosterol in fungal cell membrane
Dosage & Administration
- 150 mg single dose for Vaginal Candidiasis
- 200 mg in 1st day followed by 100 mg daily for 14 days for Oropharyngeal Candidiasis
- 200 mg in 1st day followed by 100 mg daily for 14-30 days for Oesophageal Candidiasis
- 150 mg weekly for 4-6 weeks for Tinea Infections
- 50 mg daily for 20 days for Kerion
- 400 mg as a single dose for Pityriasis Versicolor
- 150 mg weekly for 12 months for Onychomycosis
- Orally or by IV infusion, 400 mg on first day then 200-400 mg daily for Cryptococcal Infections
- Orally or by IV infusion 200 mg daily for Prevention of Cryptococcal Meningitis
- Orally or by IV infusion, 50-400 mg daily for Prevention of Fungal Infections in Immunocompromised Patients
Interaction
- Increased prothrombin time after warfarin administration
- Prolonged serum half-life of oral sulphonylureas
- Increased plasma concentrations when co-administered with hydrochlorothiazide
- Potential increase in phenytoin levels
- Potential decrease in fluconazole AUC and shorter half-life when co-administered with rifampicin
Contraindications
- Hypersensitivity to Fluconazole or related triazole compounds

Pregnancy & Lactation
- Adverse fetal effects seen in animals at high doses
- Not recommended for use in pregnancy or lactation unless with adequate contraception
Precautions & Warnings
- Abnormalities in hepatic, renal, haematological and biochemical function tests observed
- Rare cases of severe cutaneous reactions
- Use not recommended during lactation
- Unlikely to impair ability to drive or use machinery
Use in Special Populations
- Normal dose recommended for elderly with no evidence of renal impairment
- Excreted predominantly in urine, dosage adjustments required for renal impairment
Overdose Effects
- Supportive and symptomatic treatment indicated
- Forced volume diuresis may increase elimination rate
- Haemodialysis decreases plasma levels by approximately 50%
