Pharmacology
Fluconazole is a potent inhibitor of fungal cytochrome P-450 dependent enzymes and essential for fungal cell membrane synthesis.
Dosage & Administration
- Dosage varies for different indications and patient groups
- Administration includes single dose, daily doses, and weekly doses
- Dosage interval modification for patients with renal impairment
Interaction
- Interaction with warfarin and prothrombin time
- Interaction with oral sulphonyl ureas
- Interaction with hydrochlorothiazide
- Interaction with phenytoin and rifampicin
- Interaction with combined oral contraceptive
- Interaction with cyclosporin, theophyline, and food/cimetidine/antacids/radiation
- Potential for drug-drug interactions with other medications
Contraindications
Fluconazole should not be used in patients with known hypersensitivity to Fluconazole or related triazole compounds.
Side Effects

- Rarely encountered adverse events such as rash and anaphylaxis
Pregnancy & Lactation
Fluconazole should not be used in pregnancy or in women of childbearing potential unless adequate contraception is employed.
Precautions & Warnings
- Observation of hepatic, renal, hematological, and biochemical function tests
- Assessment of risk-benefit ratio for patients with significant rise of liver enzymes
- Rare development of exfoliative cutaneous reactions
- Discontinuation of therapy in case of rashes in superficial or invasive/systemic fungal infections
- F-Zol found in human breast milk at concentrations similar to plasma
Use in Special Populations
- Normal dose usage in the elderly if no evidence of renal impairment
- Dosage adjustments in renal impairment based on creatinine clearance
Overdose Effects
In the event of overdosage, supportive measures and symptomatic treatment with gastric lavage if necessary may be adequate.
