Indications

Pharmacology
- Triazole antifungal agent
- Potent inhibitor of fungal cytochrome P-450 dependent enzymes
- Responsible for the synthesis of ergosterol

Interaction
- Increased prothombin time after warfarin administration
- Prolonged serum half-life of concomitantly administed oral sulphonyl ureas
- Increased plasma concentrations of fuconazole by 40% with hydrochlorothiazide
- Increased level of phenytoin
- Phenytoin AUC decrease and shorter half-life with rifampicin
- Combined oral contraceptive effects
- Slowly increase cyclosporin concentrations
- No clinically signifcant impairment of fuconazole absorption with food, cemetidine, antacids, or following total body irradiation
- Decreased plasma clearance of theophyline
Contraindications
- Hypersensitivity to Fluconazole or related triazole compounds

- Rare cases of anaphylaxis
Pregnancy & Lactation
- Adverse fetal effects in animals only at dose levels associated with maternal toxicity
- Little use during human pregnancy
- Not recommended in pregnancy or in women of child bearing potential
Precautions & Warnings
- Abnormalities of hepatic, renal, haematological and other biochemical function tests observed in some patients
- Rare instances of hepatic necrosis
- Development of severe cutaneous reactions
- Discontinuation of therapy in case of rash
- Not recommended during lactation
- Unlikely to impair a patient's ability to drive or use machinery

Overdose Effects
- Supportive measures and symptomatic treatment
- Gastric lavage if necessary
- Forced volume diuresis to increase elimination rate
- Haemodialysis to decrease plasma levels by approximately 50%
