Indications

Pharmacology
- Potent inhibitor of fungal cytochrome P-450 dependent enzymes
- Responsible for the synthesis of ergosterol
Dosage & Administration
- Administered as a single dose for Vaginal Candidiasis
- Different dosages for different types of Candidiasis and Tinea
- Orally or by IV infusion for Invasive candidal infections and cryptococcal infections
- Dosage intervals modified based on renal impairment
- Dosage recommendations for children based on weight
- Specific dosage recommendations for special populations such as elderly patients and those with renal impairment
Interaction
- Increased prothombin time after warfarin administration
- Prolongs serum half-life of concomitantly administered oral sulphonylureas
- Increased plasma concentrations when co-administered with hydrochlorothiazide
- May increase the level of phenytoin to a clinically significant degree when co-administered
- Significant change in AUC and shorter half-life when co-administered with rifampicin
- Efects on oral contraceptive hormone levels
- Potential interaction with cyclosporin
- Can be co-administered with food, cemetidine, antacids, or following total body irradiation for bone marrow transplantation without clinically significant impairment of absorption
- May result in a decrease in the mean plasma clearance of theophylline
Contraindications
- Not to be used in patients with known hypersensitivity to Fluconazole or related triazole compounds
Side Effects
- Common side effects include symptoms associated with the gastrointestinal tract
- Rarely encountered adverse events such as rash
- Rare cases of anaphylaxis reported
Pregnancy & Lactation
- Adverse fetal effects seen in animals at dose levels associated with maternal toxicity
- Not recommended for use in pregnancy or in women of childbearing potential unless adequate contraception is employed
Precautions & Warnings
- Observation of abnormalities in hepatic, renal, haematological, and other biochemical function tests in some patients, particularly those with serious underlying diseases
- Rare development of exfoliative cutaneous reactions
- Monitoring and discontinuation of therapy in case of developed rashes
- Not recommended for use during lactation
- Unlikely to impair a patient's ability to drive or use machinery

- Recommended dosage adjustments in elderly patients and those with renal impairment
Overdose Effects
- Supportive measures and symptomatic treatment recommended
- Potential use of gastric lavage if necessary
- Forced volume diuresis may increase the elimination rate
- Haemodialysis may decrease plasma levels by approximately 50%
