Indications

Pharmacology
Triazole antifungal agent, potent inhibitor of fungal cytochrome P-450 dependent enzymes
Dosage & Administration
- Adults:
- Vaginal Candidiasis: 150 mg as a single dose
- Oropharyngeal Candidiasis: 200 mg in 1st day followed by 100 mg daily for 14 days
- Oesophageal Candidiasis: 200 mg in 1st day followed by 100 mg daily for 14-30 days
- Tinea corporis/Tinea cruris/Tinea pedis/Other Tinea: 150 mg weekly for 4-6 weeks
- Kerion: 50 mg daily for 20 days
- Pityriasis versicolor: 400 mg as a single dose
- Onychomycosis: 150 mg weekly for 12 months
- Children over 1 year:
- In Superficial Candidiasis: 1-2 mg/kg daily
- In Systemic Candidiasis & Cryptococcal infection: 3-6 mg/kg daily
- Use in Specific Population: Adjustment of dosage intervals based on renal impairment, doses up to 12 mg/kg recommended for children
Interaction
Interactions with warfarin, sulphonyl ureas, hydrochlorothiazide, phenytoin, rifampicin, oral contraceptive, cyclosporin, theophyline, food, cemetidine, antacids, total body irradiation, and other medications possible
Contraindications
Hypersensitivity to Fluconazole or related triazole compounds
Side Effects
- Common:
- Nausea
- Abdominal discomfort
- Diarrhoea
- Fatulence
- Rare: Rash, Anaphylaxis
Pregnancy & Lactation
Not recommended for use unless adequate contraception is employed
Precautions & Warnings
- Abnormalities: Observed in hepatic, renal, haematological, and other biochemical function tests in some patients with serious underlying diseases
- Hepatic Necrosis: Rarely observed, assessment of risk-benefit ratio recommended
- Exfoliative Cutaneous Reactions: Rare, caution advised in patients with rash development
- Lactation: Not recommended for nursing mothers
- Driving/Use of Machinery: Unlikely to impair ability
Use in Special Populations
- Elderly: Normal dose recommended if no evidence of renal impairment
- Renal Impairment: Adjustment of dosage intervals based on creatinine clearance
Overdose Effects
Supportive and symptomatic treatment, gastric lavage if necessary, forced diuresis may increase elimination rate, haemodialysis may decrease plasma levels by approximately 50%
