Indications

Pharmacology
Triazole antifungal agent. Potent inhibitor of fungal cytochrome P-450 dependent enzymes.
Dosage & Administration
- Adults:
- Dosage varies based on condition e.g., Vaginal Candidiasis: 150 mg as a single dose
- Oropharyngeal Candidiasis: 200 mg in 1st day followed by 100 mg daily for 14 days
- Children over 1 year:
- Dosage based on condition and weight
Interaction
- May affect prothrombin time when taken with warfarin
- May impact serum half-life of concomitantly administered oral sulphonyl ureas
- Co-administration with hydrochlorothiazide can increase plasma concentrations
- May increase the level of phenytoin and cyclosporin
- May affect combined oral contraceptive efficacy
- May slow increase cyclosporin concentrations
Contraindications
Should not be used in patients with known hypersensitivity to Fluconazole or related triazole compounds
Side Effects
Common side effects include symptoms associated with the gastrointestinal tract. Rarely reported adverse events include rash and anaphylaxis.
Pregnancy & Lactation
Not recommended for use in pregnancy or in women of childbearing potential unless adequate contraception is employed
Precautions & Warnings
- Hepatic, Renal, and Biochemical Function: Abnormalities observed, particularly in patients with serious underlying diseases
- Cutaneous Reactions: Rare occurrence of severe cutaneous reactions
- Use during Lactation: Not recommended due to presence in human breast milk
- Driving/Use of Machinery: Unlikely to impair the patient's ability to drive or use machinery
Use in Special Populations
- Elderly: Normal dosage should be used, adjustments may be required in patients with renal impairment
- Renal Impairment: Dosage intervals or daily dosage should be modified based on creatinine clearance levels
Overdose Effects
Supportive and symptomatic treatment recommended. Forced volume diuresis may increase elimination rate. Haemodialysis decreases plasma levels by approximately 50%
