Mechanism of Action

  • Dexlansoprazole is a PPI that suppresses gastric acid secretion by specific inhibition of the H+/K+-ATPase in the gastric parietal cell.

Indications

  • Gastroesophageal reflux disease (GERD)
  • Erosive esophagitis
  • Helicobacter pylori eradication in combination therapy

Common Dosage Forms

  • Delayed-release capsules

Typical Dosage

  • 30 mg orally once daily for GERD maintenance therapy
  • 60 mg orally once daily for 8 weeks for healing of erosive esophagitis

Pediatric Dosage

  • Safety and efficacy in pediatric patients have not been established

Geriatric Dosage

  • No dosage adjustment is necessary in the elderly, but caution should be exercised

Side Effects

  • Common side effects include diarrhea, abdominal pain, nausea, flatulence, and headache

Contraindications

Pregnancy Category

  • Category B - Animal reproduction studies have failed to demonstrate a risk to the fetus and there are no adequate and well-controlled studies in pregnant women.
