Indications
- Treatment of acute clinical manifestations of Herpes simplex virus in immunocompromised patients
- Treatment of severe primary or non-primary genital herpes in immune competent patients
- Treatment of Varicella zoster virus infection in immunocompromised patients
- Treatment of Herpes zoster (shingles) in immune competent patients
- Treatment of Herpes simplex encephalitis
Pharmacology
Interferes with DNA synthesis and inhibits viral replication for Herpes simplex virus and Varicella zoster virus
Dosage
- 5 mg/kg every 8 hours for Herpes simplex infection
- 5 mg/kg every 8 hours for very severe Herpes zoster infection
- 10 mg/kg every 8 hours for Varicella zoster infection
- 10 mg/kg every 8 hours for Herpes simplex encephalitis
Administration
- Administered for 5-7 days in most infections and at least 10 days for Herpes simplex encephalitis
- Should be administered by slow intravenous infusion over a one-hour period
- Can be injected directly into a vein or further diluted for administration by infusion
Interaction
Co-administration of probenecid or diuretics can affect plasma levels of the drug
Contraindications
Hypersensitivity to Acyclovir or Valacyclovir

- Pruritus
- Increases in liver-related enzymes
- Local inflammatory reactions
Pregnancy & Lactation
Pregnancy category B; should be used in pregnancy only if benefits outweigh risks
Precautions & Warnings
- Intended for intravenous infusion only
- Reconstituted infusion should not be administered by mouth
- Should be given over a period of at least one hour to avoid renal tubular damage

- Pediatric use: Dose calculated on the basis of body surface area

Overdose Effects
- Elevations of serum creatinine and blood urea nitrogen
- Neurological effects like confusion, seizures, and coma
- Adequate hydration essential to reduce the possibility of crystal formation in urine
