Dosage & Administration
- Adults: 10-20 mg, every 6-8 hours daily (maximum of 80 mg daily)

- Treatment of acute nausea and vomiting
- Use in Parkinson's disease for dopamine-agonist induced nausea and vomiting

Pharmacology
- Dopamine antagonist
- Blocks dopamine receptors in the CTZ and stomach
- Restores normal motility and tone of the upper gastrointestinal tract
- Weak penetration across the blood-brain barrier
Interaction
- Antagonized by anticholinergic drugs
- Not to be given simultaneously with antacids and antisecretory drugs
- Metabolized through CYP3A4 pathway
- Potential influence on absorption of concomitantly orally administered drugs
Contraindications
- Hypersensitivity to the drug
- Neonates
- Gastrointestinal hemorrhage, mechanical obstruction, or perforation
- Prolactin releasing pituitary tumor
Side Effects
- Rare occurrence of transient intestinal cramps
- Rare extrapyramidal phenomena, especially in children
- Possible induction of hyperprolactinaemia
- Rare allergic reactions such as rash and urticaria
Pregnancy & Lactation
- Caution in use during the first trimester of pregnancy
- Excretion in breast milk, caution advised for nursing mothers
Precautions & Warnings
- Caution in use with children due to risk of extra-pyramidal reactions
- Use with caution in patients with hepatic impairment

Overdose Effects
- Symptoms include drowsiness, disorientation, and extrapyramidal reactions
