Description
- Dopamine antagonist medication for treating nausea and vomiting
- Gastroprokinetic action on the motility of the gastrointestinal tract
Indications
- Epigastric sense of fullness

Pharmacology
- Dopamine antagonist
- Blocks dopamine receptors in the ChemoreceptorTrigger Zone (CTZ) and stomach
- Restores normal motility and tone of the upper gastrointestinal tract
- Facilitates gastric emptying
- Enhances antral and duodenal peristalsis
- Regulates contraction of the pylorus
- Increases esophageal peristalsis and lower esophageal sphincter pressure
Dosage & Administration
- Should be taken 15-30 minutes before meals
- Usual recommended oral dose for adults: 10-20 mg every 6-8 hours daily
- Maximum dose of 80 mg daily
- Dosage for children based on body weight and condition
Interactions
- Antagonized by anticholinergic drugs
- Lower oral bioavailability when given with antacids and antisecretory drugs
- Main metabolic pathway through CYP3A4
- Potential interaction with drugs that inhibit CYP3A4
- May influence absorption of concomitantly orally administered drugs
Contraindications
- Known hypersensitivity to the drug
- Neonates
- Gastrointestinal hemorrhage, mechanical obstruction, or perforation
