Pharmacology

  • Dopamine antagonist
  • Blocks dopamine receptors in the ChemoreceptorTrigger Zone (CTZ) and stomach
  • Restores normal motility and tone of the upper gastrointestinal tract
  • Facilitates gastric emptying
  • Enhances antral and duodenal peristalsis
  • Regulates contraction of the pylorus
  • Increases esophageal peristalsis and lower esophageal sphincter pressure

Dosage & Administration

  • Should be taken 15-30 minutes before meals
  • Usual recommended oral dose for adults: 10-20 mg every 6-8 hours daily
  • Maximum dose of 80 mg daily
  • Dosage for children based on body weight and condition

Interactions

  • Antagonized by anticholinergic drugs
  • Lower oral bioavailability when given with antacids and antisecretory drugs
  • Main metabolic pathway through CYP3A4
  • Potential interaction with drugs that inhibit CYP3A4
  • May influence absorption of concomitantly orally administered drugs
