Indications

  • Dyspeptic symptom complex

Pharmacology

  • Blocks dopamine receptors in the Chemoreceptor Trigger Zone (CTZ) and stomach
  • Gastroprokinetic action
  • Restores normal motility and tone of the upper gastrointestinal tract
  • Facilitates gastric emptying

Dosage & Administration

  • Adults oral dose: 10-20 mg every 6-8 hours
  • Children oral dose: 2-4 ml suspension/10 kg body weight every 6-8 hours
  • Adults rectal dose: 30-60 mg every 4-8 hours

Interaction

  • Anticholinergic drugs antagonize antidyspeptic effect
  • Simultaneous use with antacids and antisecretory drugs lowers oral bioavailability
  • Inhibition of CYP3A4 enzyme

Contraindications

  • Known hypersensitivity to the drug
  • Neonates
  • Gastrointestinal hemorrhage, mechanical obstruction, or perforation

Side Effects

  • Transient intestinal cramps
  • Extrapyramidal phenomena
  • Increase in plasma prolactin level

Pregnancy & Lactation

  • Use during first trimester justified by anticipated therapeutic benefit
  • Excretion in breast milk
