Description
- A medication used for the treatment of dyspeptic symptom complex, acute nausea and vomiting, Parkinson's disease, and as an aid in radiological studies. It is a dopamine antagonist with gastroprokinetic action.

Pharmacology
- Dopamine antagonist
- Blocks dopamine receptors in the CTZ and stomach
- Restores normal motility and tone of upper gastrointestinal tract
- Enhances antral and duodenal peristalsis
- Increases esophageal peristalsis and lower esophageal sphincter pressure
Interaction
- Antagonized by anticholinergic drugs
- Lowered oral bioavailability when given with antacids or antisecretory drugs
- Metabolized through CYP3A4 pathway
- Potential effects on absorption of concomitantly orally administered drugs
Contraindications
- Known hypersensitivity to the drug
- Neonates
- Gastrointestinal hemorrhage
- Mechanical obstruction or perforation
- Prolactin releasing pituitary tumor
Side Effects
- Transient intestinal cramps
- Extrapyramidal phenomena (rare)
- Increase in plasma prolactin level
- Rare allergic reactions such as rash and urticaria
Pregnancy & Lactation
- Use only during the first trimester if justified by therapeutic benefit
- Excreted in breast milk at lower concentrations than in plasma
- Not recommended during nursing unless benefits outweigh risks
