Pharmacology
- Dopamine antagonist
- Blocks dopamine receptors in the ChemoreceptorTrigger Zone (CTZ) and stomach
- Gastroprokinetic action
- Restores normal motility and tone of the upper gastrointestinal tract
- Facilitates gastric emptying
- Enhances antral and duodenal peristalsis
- Regulates contraction of the pylorus
- Increases esophageal peristalsis and lower esophageal sphincter pressure
Dosage administration
- Recommended to be taken 15-30 minutes before meals
- Adults: 10-20 mg every 6-8 hours daily
- Children: 2-4 ml suspension/10 kg body weight every 6-8 hours daily
- Maximum dose of 80 mg daily for adults
- Maximum period of treatment is 12 weeks
- Rectal administration for adults and children
Interaction
- Concomitant administration of anticholinergic drugs may antagonize the antidyspeptic effect
- Antacids and antisecretory drugs should not be given simultaneously
- Metabolic pathway through CYP3A4
- Inhibition by drugs such as azole antifungals, macrolide antibiotics, HIV protease inhibitors
- Potential influence on absorption of concomitantly orally administered drugs
