Description
Perion 10 mg Tablet is a dopamine antagonist medication which is used to treat nausea and vomiting. It helps relieve dyspeptic symptom complex, acute nausea and vomiting, and Parkinson's disease-induced nausea and vomiting. It also aids in speeding barium transit in follow-through radiological studies.

Dosage Instructions
- Adults: 10-20 mg (1-2 tablets or 10-20 ml suspension), every 6-8 hours daily
- Children: 2-4 ml suspension/10 kg body weight or 0.4-0.8 ml paediatric drops/10 kg body weight, every 6-8 hours daily

Pharmacology
Dopamine antagonist that blocks dopamine receptors in the Chemoreceptor Trigger Zone (CTZ) and stomach. Gastroprokinetic action based on blocking effect of dopamine receptors influencing gastrointestinal motility.
Mechanism of Action
- Restores normal motility and tone of upper gastrointestinal tract
- Facilitates gastric emptying
- Enhances antral and duodenal peristalsis
- Regulates contraction of the pylorus
- Increases esophageal peristalsis and lower esophageal sphincter pressure
Interaction with Other Drugs
- Anticholinergic drugs may antagonize the antidyspeptic effect of domperidone
- Simultaneous administration with antacids and antisecretory drugs should be avoided
- Metabolized through CYP3A4, so concomitant use of CYP3A4 inhibitors may result in increased plasma levels
- Potential influence on absorption of concomitantly orally administered drugs
Contraindications
- Known hypersensitivity to domperidone
- Neonates
- Gastrointestinal hemorrhage, mechanical obstruction, perforation
- Prolactin releasing pituitary tumor (prolactinoma)
Side Effects
- Transient intestinal cramps
- Rare extrapyramidal phenomena
- Increase in plasma prolactin level
- Rare allergic reactions such as rash and urticaria
Pregnancy & Lactation
- Caution advised during the first trimester of pregnancy
- Excreted in breast milk of lactating rats
- Unknown potential harm to newborns
- Effect on ability to drive and use machines: No effect on mental alertness
Precautions & Warnings
- Use with caution in children due to risk of extra-pyramidal reactions
- Use with caution in patients with hepatic impairment
Special Population Uses
- Use in infants with caution under close medical supervision
- Caution in patients with liver disorders
- Dosage adjustment may be required in patients with severe renal insufficiency
Overdose Effects
- Symptoms include drowsiness, disorientation, extrapyramidal reactions
