Description
- Dopamine antagonist used for treating dyspeptic symptom complex, acute nausea and vomiting, Parkinson's disease, and speeding barium transit in radiological studies.
Dosage & Administration
- Should be taken 15-30 minutes before meals
- Adults: 10-20 mg every 6-8 hours daily, maximum 80 mg daily
- Children: 2-4 ml suspension/10 kg body weight every 6-8 hours daily
- Rectal administration: Adults 30-60 mg every 4-8 hours, Children maximum 30 mg for those weighing 10-25 kg

Pharmacology
- Dopamine antagonist
- Blocks dopamine receptors in Chemoreceptor Trigger Zone and stomach
- Restores normal motility and tone of upper gastrointestinal tract
- Facilitates gastric emptying
- Enhances antral and duodenal peristalsis
- Increases esophageal peristalsis
- Regulates contraction of the pylorus
Interaction
- Contraindicated with anticholinergic drugs
- Do not administer simultaneously with antacids and antisecretory drugs
- Metabolized through CYP3A4
- May interact with CYP3A4 inhibitors like azole antifungals, macrolide antibiotics, HIV protease inhibitors
Contraindications
- Known hypersensitivity to the drug
- Neonates
- Gastrointestinal hemorrhage, mechanical obstruction, perforation
- Prolactin releasing pituitary tumor (prolactinoma)
Side Effects
- Rare intestinal cramps
- Rare extrapyramidal phenomena in young children and adults
- Possible increase in plasma prolactin level
- Rare allergic reactions like rash and urticaria
Pregnancy & Lactation
- Use in first trimester if justified
- Excreted in breast milk
- Not recommended for nursing mothers unless benefits outweigh potential risk
