Medicine Details

Indications

  • Gastric and duodenal ulcer
  • NSAID-associated duodenal and gastric ulcer
  • As prophylaxis in patients with a history of NSAID-associated duodenal and gastric ulcer
  • Gastro-esophageal reflux disease
  • Long-term management of acid reflux disease
  • Acid-related dyspepsia
  • Severe ulcerating reflux esophagitis
  • Prophylaxis of acid aspiration during general anesthesia
  • Zollinger-Ellison syndrome
  • Helicobacter pylori-induced peptic ulcer

Pharmacology

Inhibitor of gastric acid secretion, onset of antisecretory effect within one hour, maximum effect within two hours, inhibition lasts up to 72 hours

Dosage

  • Oral administration
  • 20mg once daily for benign gastric and duodenal ulcer
  • 10-20mg daily for long-term acid reflux disease management
  • 40mg on preceding evening for prophylaxis of acid aspiration
  • 60mg once daily for Zollinger-Ellison syndrome
  • 20mg twice daily for Helicobacter pylori eradication regimen in peptic ulcer disease
  • IV Injection: 40mg slowly, IV infusion: 20-30 minutes or more

Administration

  • Direction for use of IV Injection
  • Direction for use of IV Infusion
  • IV injection should be given slowly over a period of at least 2 to 5 minutes at a maximum rate of 4 ml/minute

Interaction

  • Reduced absorption of ketoconazole during Omegut treatment
  • Metabolised in the liver through cytochrome P450
  • Possible delay in elimination of diazepam, phenytoin, and warfarin

Contraindications

Known hypersensitivity to Omeprazole, suspicion of malignancy when gastric ulcer is suspected

Side Effects

  • Mild and reversible adverse reactions
  • Skin rash, urticaria, pruritus
  • Photosensitivity, bullous eruption, erythema multiforme
  • Diarrhea, headache, constipation, nausea/vomiting
  • Dry mouth, stomatitis, candidiasis
  • Dizziness, lightheadedness, somnolence
  • Arthritic and myalgic symptoms
  • Blurred vision, taste disturbance, peripheral edema
  • Increases in liver enzymes

Pregnancy & Lactation

  • Results indicate no adverse effects on pregnancy or fetus/child health
  • Use during pregnancy is permissible
  • No information available on passage into breast milk or effects on neonate
  • Breastfeeding should be discontinued if Omeprazole use is essential
  • Safety and effectiveness not established in pediatric patients under 18 years

Precautions & Warnings

  • Avoid concomitant use of clopidogrel and Omegut
  • Increased risk for osteoporosis-related fractures with PPI therapy
  • Atrophic gastritis noted occasionally with long-term use
  • Concomitant use with methotrexate may lead to toxicities
