Indications
- Gastric and duodenal ulcer
- NSAID-associated duodenal and gastric ulcer
- Prophylaxis in patients with a history of NSAID-associated duodenal and gastric ulcer
- Gastro-esophageal reflux disease
- Long-term management of acid reflux disease
- Acid-related dyspepsia
- Severe ulcerating reflux esophagitis
- Prophylaxis of acid aspiration during general anesthesia
- Zollinger-Ellison syndrome
- Helicobacter pylori-induced peptic ulcer
Pharmacology
- Inhibitor of gastric acid secretion
- Actions on hydrogen-potassium-adenosine triphosphatase enzyme system
- Onset of antisecretory effect within one hour
- Maximum effect occurring within two hours
- Inhibition of secretion lasting up to 72 hours
Dosage
- 20 mg once daily for benign gastric and duodenal ulcer
- 40 mg daily for severe or recurrent cases
- 10-20 mg daily for NSAID-associated duodenal or gastric ulcer prophylaxis
- 20 mg once daily for gastro-esophageal reflux disease
- 40 mg once daily for 8 weeks in refractory cases of gastro-esophageal reflux disease
- 10-20 mg daily for long-term management of acid reflux disease
- 40 mg for prophylaxis of acid aspiration
- 60 mg once daily for Zollinger-Ellison syndrome
Administration
- IV injection for prophylaxis of acid aspiration and severe conditions
- IV infusion for prolonged administration
- Directions for use of IV injection and infusion
Interaction
- Effect on the absorption of ketoconazole
- Metabolism through cytochrome P450
- Monitoring and dose adjustment for warfarin and phenytoin
- No change in blood concentration of phenytoin and coagulation time with concomitant treatment
- Plasma concentration changes with clarithromycin
- No evidence of interaction with certain drugs
- Effect on digoxin bioavailability
Contraindications
- Hypersensitivity to omeprazole
- Suspected gastric ulcer without excluding malignancy
Side Effects

- Photosensitivity and bullous eruption
- Gastrointestinal reactions like diarrhea, constipation, and nausea/vomiting
- Neurological symptoms like headache, dizziness, and paraesthesia
Pregnancy & Lactation
- No adverse effects on pregnancy or fetus/newborn child reported
- Use during pregnancy with no known harmful effects
- No information on passage into breast milk
- Discontinuation of breastfeeding if essential
Precautions & Warnings
- Avoid concomitant use of clopidogrel
- Possible risk of osteoporosis-related fractures with PPI therapy
